Wind Noise From Drivers Window

I have another problem related to the door window. The window gets loose all the time. I had to take the window out twice to tighten the front plastic guide that attaches to the window. It seems like it's a bad design. When you tighten the screw the end of the plastic guide mushrooms and tightens against the glass.  The hardware is new. I was thinking of replacing the screw with a bolt and use a nylon lock nut.Any suggestions?  Thanks

 If the screw is coming loose, I suppose you could replace it with a bolt but I would be wondering why it is coming loose as that is not normal.
