Will a 5/16" primary fuel line provide enough fuel for a mildly modified single 4 barrel (holley 850) 440? I am expecting the engine to produce 450 to 500 HP. Should I go to a 3/8" line? Also does anyone know how to make the expanded flare on the end of the fuel line so a rubber hose can be connected to the line to eliminate leaking?

I would use 3/8's.   Not sure about the flare...I just double clamp them

I use a double flare & swell the tube with no sharp edges , slide the hose over & clamp it

Most places like Ace, Home Depot, NAPA,  etc carry the tools required to do it. Their cheap to buy and easy to use.

unless you are running the quarter mile full throttle, I would think you 5/16 would be fine.
As long as you got a good fuel pump,and clean fuel filter, you should be ok.
A fuel pressure gauge in the car can really help you know for sure.

Sometimes the fuel sending unit sock filter in the gas tank can clog up.
